Exploring video and eDNA metabarcoding methods to assess oyster aquaculture cages as fish habitat

چکیده

Multi-tiered oyster aquaculture cages may provide habitat for fish assemblages similar to natural structured seafloor. Methods were developed assess associated with gear and boulder using underwater video census combined environmental DNA (eDNA) metabarcoding. Action cameras mounted on 3 at a commercial eastern Crassostrea virginica farm (‘cage’) among boulders rock reef (‘boulder’) from June August 2017 in Long Island Sound, USA. Interval continuous recording strategies tested. During interval recording, collected 8 min segments hourly 07:00 19:00 h only. Continuous was also 2-3 boulders. Data loggers recorded light intensity current speed. Seawater eDNA metabarcoding the farm. MaxN measurements of abundance calculated video, 7 species observed. Black sea bass Centropristis striata , cunner Tautogolabrus adspersus scup Stenotomus chrysops tautog Tautoga onitis most abundant observed both cage videos. In black bass, scup, more frequently higher farm, while within reef. detected 42 Six methods. Applied tandem, provided comprehensive approach describing difficult sample habitats.

Fish community assessment with eDNA metabarcoding: effects of sampling design and bioinformatic filtering

Species richness is a metric of biodiversity that represents the number of species present in a community. Traditional fisheries assessments that rely on capture of organisms often underestimate true species richness. Environmental DNA (eDNA) metabarcoding is an alternative tool that infers species richness by collecting and sequencing DNA present in the ecosystem. Our objective was to determin...

Biomonitoring of marine vertebrates in Monterey Bay using eDNA metabarcoding

Molecular analysis of environmental DNA (eDNA) can be used to assess vertebrate biodiversity in aquatic systems, but limited work has applied eDNA technologies to marine waters. Further, there is limited understanding of the spatial distribution of vertebrate eDNA in marine waters. Here, we use an eDNA metabarcoding approach to target and amplify a hypervariable region of the mitochondrial 12S ...

Aquaculture and Fish Health

Introduction Fish has an enormous value as the most frequent source of animal protein for the major part of the humans, especially in countries during rapid development in Asia and Africa. All fish species are a part of the local ecology and as such has a biological and environmental importance. Fishing also gives a great pleasure to sport fishermen around the world and at the same time contrib...

Transgenic fish and aquaculture

Transgenic fish species can be routinely produced by transferring foreign DNA into developing embryos via microinjection or electroporation. This technology offers an excellent opportunity for modifying or improving the genetic traits of commercially important Fishes, mollusks, and crustaceans for aquaculture. Studies have shown that administration of recombinant fish or mammalian growth hormon...
