Exploring video and eDNA metabarcoding methods to assess oyster aquaculture cages as fish habitat

Multi-tiered oyster aquaculture cages may provide habitat for fish assemblages similar to natural structured seafloor. Methods were developed assess associated with gear and boulder using underwater video census combined environmental DNA (eDNA) metabarcoding. Action cameras mounted on 3 at a commercial eastern Crassostrea virginica farm (‘cage’) among boulders rock reef (‘boulder’) from June August 2017 in Long Island Sound, USA. Interval continuous recording strategies tested. During interval recording, collected 8 min segments hourly 07:00 19:00 h only. Continuous was also 2-3 boulders.
